Phosphate Buffered Saline (PBS) is a sterile, isotonic buffer solution composed of sodium chloride, sodium phosphate, and, in some formulations, potassium salts. It is widely used in biological and biochemical research to maintain physiological pH and osmolarity, providing a stable environment for peptides, proteins, and cells. PBS is not a bioactive compound but functions as a carrier and stabilisation medium, commonly used for dilution, washing, and reconstitution in laboratory and research settings. Its compatibility with biological systems makes it a standard solution for maintaining structural integrity and preventing osmotic stress.
Mechanism of Action
PBS functions as a buffered isotonic solution, maintaining a stable hydrogen ion concentration through the phosphate buffer system. This prevents fluctuations in pH that could otherwise denature peptides or alter protein structure. The sodium chloride component ensures osmotic balance, preventing cellular swelling or shrinkage, while phosphate ions stabilise the environment by resisting changes in acidity. PBS does not interact directly with biological targets but instead preserves the structural and functional integrity of biomolecules.
