Oxytocin

Evidence profile: Controlled human evidence

Oxytocin is a cyclic nonapeptide hormone and neuropeptide produced in the hypothalamus and released by the posterior pituitary. It plays a central role in social bonding, emotional regulation, reproduction, childbirth, and lactation, earning the designation as the ‘bonding’ or ‘social’ hormone. Clinically, synthetic oxytocin (e.g., Pitocin, Syntocinon) is FDA-approved for labor induction and postpartum hemorrhage control. Beyond obstetrics, intranasal and experimental uses are being investigated for neuropsychiatric conditions including autism spectrum disorder, anxiety, PTSD, and social cognition disorders.

Mechanism of Action

Oxytocin binds to oxytocin receptors (OXTR), which are G-protein coupled receptors expressed in uterine smooth muscle, mammary tissue, and the central nervous system. In uterine tissue, receptor activation increases intracellular calcium, triggering myometrial contractions and prostaglandin release. In the brain, oxytocin modulates limbic system activity, influencing social behavior, emotional processing, and stress responses.
